SRX10191935: ONT PromethION WGS of Sandy the Desert Dingo: skin biopsy

Design: DNA (1 g) was prepared for ONT sequencing using the 1D genomic DNA by ligation kit (SQK-LSK109, ONT) according to the standard protocol. Long fragment buffer was used for the final elution to exclude fragments shorter than 1000 bp. In total, 119 ng of adapted DNA was loaded onto a FLO-PRO002 PromethION flow cell and run on an ONT PromethION sequencing device using MinKNOW (18.08.2) with MinKNOW core (v1. 14.2). Base-calling was performed after sequencing with the GPU-enabled guppy basecaller (v3.0.3; high accuracy flip-flop model; config dna_r9.4.1_450bps_hac.cfg). Sequencing was conducted at Kinghorn Centre for Clinical Genomics at the Garvan Institute of Medical Research, Sydney, Australia.
